冠心合剂对心梗后大鼠VEGF表达的影响 被引量:2

摘要 目的:观察冠心合剂对急性心肌梗死(AMI)大鼠血管内皮生长因子(Vascular endothelial growth fac-tor,VEGF)表达的影响。方法:结扎冠状动脉左前降支法建立大鼠急性心肌梗死模型,观察冠心合剂对心梗后大鼠血清VEGF浓度、心肌VEGF蛋白表达及梗死面积的影响。结果:冠心合剂治疗组及阳性对照组血清VEGF浓度及心肌VEGF蛋白的表达增加,高于模型组和假手术组(P<0.05);随冠心合剂剂量的增加,梗死面积逐渐减小;冠心合剂大剂量组疗效与西药对照组无明显差异(P>0.05)。结论:冠心合剂可能通过促进心肌梗死大鼠心肌VEGF蛋白表达、分泌,从而促进梗死心肌血管新生。 Objective :To investigate the effect of Guanxin Decoction on myocardial angiogenesis and the expression of VEGF in rats with acute myocardial infarction. Methods: Left anterior descending coronary arteries (LAD) have been ligated in SD rats after anesthetized by 2.5 % pentobarbital sodium. The level of VEGF in blood serum was measured 2 weeks after the experiment by ELISA respectively. The heart specimen were fixed, sectioned and colorized by means of HE staining and staining to observe VEGF through microscope. Results: Guanxin Decoction - treatment group and positive control group were higher than the model group and the sham - operation group in the expression of VEGF protein ( P 〈 0.05 ). The level of VEGF in blood serum increased correspondingly with the increase of the dose of Guanxin Decoction, while the myocardial infarction scope reducing. The curative effect of the high - dosage Guanxin Decoction is similar with bFGF and heparin( P 〉 0.05 ). Conclusion: The mechanism of AMI rat angiagenesis which is prorooted by Guanxin Decoction may be associated with the increase of expression and excretion of VEGF in cardiac myocytes.
