正交设计优选玉米须多糖酶解提取工艺 被引量:3

摘要 目的选择玉米须多糖的最佳提取工艺。方法①以正交设计实验优化提取工艺。②用浓硫酸-苯酚比色法于490nm波长处测定不同工艺提取玉米须多糖的含量。结果不同酶酶解提取的多糖含量差别不大,但是最佳酶解时间、酶解温度、酶解浓度都不一样。纤维素酶酶解提取玉米须多糖最佳提取工艺:酶解时间40min、酶解温度80℃、酶解浓度1.0%;木瓜蛋白酶酶解提取玉米须多糖结果分析:不同酶解时间间有统计学差异(P=0.033),最佳提取工艺:酶解时间120min,酶解温度40℃、酶解浓度1.5%。结论这2种优选工艺可作为玉米须多糖提取方法,也可为保健品的开发奠定一定的基础。 Objective To screen the best extraction conditions of the polysaccharides in Corn Silk.Methods ①The extraction technology was optimized by the orthogonal test.②The content of polysaccharides in Corn Silk was determined using different extraction methods with concentrated sulfuric acid-phenol colorimetric at 490 nm.Results The polysaccharide content showed no significant change by different enzymolysis extraction technologies,but the best hydrolysis time,hydrolysis temperature,enzyme concentration were different.The best cellulase enzymolysis conditions for the polysaccharides in Corn Silk were as follows:hydrolysis time 40 min,hydrolysis temperature 80 ℃,enzyme concentration 1.0%.The best caroid enzymolysis conditions for the polysaccharides in Corn Silk were:hydrolysis time 120 min,hydrolysis temperature 40 ℃,enzyme concentration 1.5%.Conclusion This optimized technologies can be used to extract the polysaccharides from Corn Silk,and also lay a foundation for the development of health care products.
