扶正解毒含药血清对染镍细胞中信号转导通路的影响 被引量:1

Effect of Fuzheng Jiedu Decoction on Signaling Pathway Induced by Nickel in Human Bronchus Epithelial cells

摘要 目的通过结晶硫化镍在支气管上皮细胞系(16 HBE)中磷酸化氨基末端蛋白激酶(P-JNK)和P38的表达,探讨镍致癌的分子机制和中草药对职业性肿瘤的防治作用。方法用SDS聚丙烯酰胺凝胶电泳和蛋白免疫印迹杂交(Western blot)法检测16 HBE细胞中P-JNK和P38的表达。结果硫化镍能明显激活P-JNK和P38的表达,硫化镍浓度越高,磷酸化P-JNK和P38的表达值越高,而且存在剂量-效应关系,染镍组与对照组比较,差异有统计学意义(P<0.05、P<0.01)。结论结晶型硫化镍激活16 HBE细胞中P-JNK和P38的表达,可能是镍致癌的分子机制之一,扶正解毒含药血清对职业性肿瘤有一定的防治作用。 [Objective]to explore the molecular mechanism of nickel carcinogenesis and preventive and therapeutic effect of Chinese herbal medicine on occupational cancer,through the study of nickel purite on expression of P-JNK and P38 in human bronchus epithelial(16 HBE) cells.[Methods]SDS polyacrylamide gel electrophoresis and Western blot hybridization method were adopted to detect the expression of P-JNK and P38.[Results]Nickel sulfide could significantly activate P-JNK and P38expression.The expression value increased with the increasing doses of nickel sulfide,with dose-effect relationship.Compared nickel sulfide group with control group,the difference was significant(P〈0.05,P〈0.01).[Conclusion]Nickel sulfide can induce the expression of P-JNK and P38 in 16HBE cells which may be the one of molecular mechanisms of cancer induced by nickel.Fuzheng Jiedu decoction has a certain preventive effect against occupational cancer.
