摘要
以水稻幼苗的叶片为材料,用0.2mol/LpH6.2的磷酸缓冲液(含1mmol/LEDTA-2Na,5%的水溶性PVP)作为谷胱甘肽过氧化物酶(GSH-Px)的提取介质,偏磷酸为酶促反应的蛋白质沉淀剂,二硫代对二硝基苯甲酸(DTNB)与GSH显色反应3min,在412nm测定酶管和非酶管的OD值,以测定谷胱甘肽过氧化物酶的活性。
In the present experiment, 0.2 M PBS (pH 6.2) containing 1 mM EDTA 2Na and 5% PVP was shown to be suitable for the extraction of GSH Px in the leaves of rice seedlings, and HPO 3 was a good protein precipitant in enzyme reactions. The activity of GSH Px could be calculated by determining the optical density (OD) of the enzyme tube and the nonenzyme tube at 412 nm of wave length after GSH had reacted with DTNB for 5 minutes.
