摘要 运用Mannich反应,将褪黑激素与BSA联结起来制备完全抗原,用UV-260紫外仪测定MLT与BSA的联结比为12∶1。用此抗原免疫新西兰大耳兔5只,经过6次免疫后,发现抗体效价平均1∶10000(ELISA)。将抗血清经Na2SO4沉淀、Sephadex-G-200过柱层析、收集纯化抗体,用分光光度计721测其蛋白浓度。经5-羟色胺等阻断试验发现抗体特异性较高。 Melatonin complete antigen is successfully synthesized through mannich formaldehyde condensation reaction with molar ratio of melatonin to BSA 12∶1. Five New Zealand Rabbits were immunized against the antigen,with the average of antiserum titers of 1∶10000. The antiserum were treated with Na 2SO 4 and Sephadex-G-200. The purified antibody is collected and with UV-721 table Spectrophotometrically,with no cross reactions for 5 methoxy tryptophan.
