中空纤维离心超滤-HPLC法分析藿香正气水中的橙皮苷 被引量:5

Separation and determination of hesperidin in Huoxiangzhengqi water by hollow fiber ultrafiltration-high performance liquid chromatography

摘要 目的:建立分析含有高分子杂质的中草药中极性物质的中空纤维分离离心超滤前处理方法,并用于藿香正气水中橙皮苷的分析。方法:试验采用新型中空纤维微量离心超滤装置(HFCUF)纯化样品溶液,在离心力作用下,利用中空纤维超滤膜除去样品溶液中的大分子,提高了色谱柱的使用寿命、分析的重现性和准确性。分析方法Promosil C18色谱柱(4.6 mm×150 mm,5μm),流动相甲醇-0.5%的乙酸水溶液(35∶65),流速1 mL.min-1,检测波长283 nm,柱温30℃。结果:橙皮苷在4.69~150 mg.L-1线性关系良好(r=0.999 7),回收率103.0%,RSD 1.9%。结论:该法简便快速、结果准确,为中药中极性活性成分的测定提供了新的、简单廉价的前处理手段。 A simple hollow fiber based centrifuge uhrafihration pretreatment procedure has been developed for the analysis of active components with high polarity in Chinese traditional and herbal drugs which usually contain macromolecule impurities. The procedure combined with HPLC was applied to the determination of hesperidin in Huoxiangzhengqi water. Sample solutions were purified by our patent hollow fiber centrifuge ultrafihration device. Under the effect of the centrifgual force, micromolecules were removed from solution samples, thus it increased the service life of the column. The accuracy and repeatability of this method have also been improvedl The separation was carried out on a Promosil Clseolumn (4. 6 mm× 150 mm, 5 μm) with methanol -0. 5% acetic acid solution (35: 65) as mobile phase at a flow rate of 1.0 mL min -1. The detector wavelength was 283 nm and the column temperature was 30 ℃. A good linear relation was obtained in the range of 4. 69 - 150 mg L-1 (r =0. 999 7) and the average recovery was 103.0% with RSD of 1.9%. This method is simple, rapid and accurate, and it provides a simple and cheap uhrafihration means for the analysis of the polar components in Chinese traditional and herbal drugs.
