摘要
目的:观察芪参益气滴丸对高脂血家兔血小板胞内游离钙浓度([Ca2+]i)的影响,并探讨芪参益气滴丸抗血小板活化的机制。方法:16只大耳白兔随机分为正常对照组和高脂喂养组,定期监测血TC值,待正常组与高脂组血脂稳定且有显著差异时,颈总动脉取血,采用高脂血浆孵育正常血小板并用体外给药的方法,运用多功能读板机检测并计算血小板胞浆内[Ca2+]i浓度的变化。结果:芪参益气组[Ca2+]i为(253.26±153.00)mmol/L较模型组[Ca2+]i(401.68±126.59)mmol/L明显下降(经平方数值变换,t=-9.691 7,P<0.01),与阿司匹林组[Ca2+]i(214.62±69.25)mmol/L比较,无显著性差异(P>0.05)。结论:芪参益气滴丸对体外高脂血浆孵育的血小板活化所致[Ca2+]i的升高有明显抑制作用。
Objective : To observe the effect of Drop Pill of Qi-shen on the saturation of free calcium in intracellular of platelet in the rabbit with hyperlipemia. Method: 16 albino rabbits with big ears were divided into normal control group and treatment group randomly. The rabbits in treatment group were fed up with rich fat food. Blood TC was monitored regularly. When TC was unchanged sometime and the two groups showed significant difference, we drew blood from arteria carotis communis. Then we incubated orthoplastocyte with rich fat blood plasma and administrated in vitro. The saturation of [ Ca^2+ ] i in platelet kytoplasm was detected and calculated with multifunctional reading machine. Result : The saturation of [ Ca^2+ ] i in treatment group was ( 253.26 ± 153.0)mmol/L, lower remarkably than (401.68 ± 126.59) mmol/L in control group (t = - 9. 691 7, P 〈 0. 01 ). But no signifieant difference showed between treatment group and aspirin group (214. 62± 69.25 mmol/ L) ( P 〉 0. 05 ). Conclusion : Drop Pill of Qi-shen can inhibit the increase of [ Ca^2 + ] i markedly caused by platelet activation which is incubated with rich fat blood plasma in vitro.
