核转录因子-κB抑制剂对慢性阻塞性肺疾病气道、肺血管重构的防治作用 被引量:6

摘要 目的:探讨核转录因子-κB(NF-κB)抑制剂吡咯烷二硫代氨基甲酸盐(PDTC)对慢性阻塞性肺疾病(COPD)气道、肺血管重构的影响。方法:将48只雄性SD大鼠随机分为8组。模型制作完成后测定气道阻力、肺血流动力学和右心室肥厚指数,观测气道炎症,气道重构以及肺小动脉的病理改变,免疫组化法检测肺组织中NF-κB蛋白的表达。结果:(1)C、D组RVSP、mPAP、RV/LV+S、MT%、MA%、肌化型动脉百分比高于A组。D组肺小动脉WT%、WA%比值增大。C1、D1组RVSP、mPAP、MT%、MA%、肌化型动脉百分比较C、D组降低。D1组的RV/LV+S、WT%、WA%值较D组降低。(2)C、D组NF-κB蛋白表达强于A组,C1、D1组较C、D组NF-κB蛋白含量下降。NF-κB表达与气道、血管重构指标均呈正相关。结论:COPD早期已出现肺血流动力学变化,随疾病进展逐渐出现进行性加重的肺血管、气道重构,NF-κB的表达与气道、血管重构有一定的相关性,PDTC可抑制NF-κB的表达,部分改善气道、肺动脉重构。
