摘要
目的研究硫酸锌对镉染毒至脐静脉内皮细胞(HUVECS)损伤的保护作用及其机制。方法人脐静脉内皮细胞体外培养,随机分为正常对照组、模型组、硫酸锌组,分别以空白血清、Cdcl2血清、Cdcl2及硫酸锌血清培养48h,收集培养液,硝酸还原酶法测定NO浓度;免疫学酶联免疫吸附法测定细胞间黏附因子(sICAM-1)及细胞色素C(CytC)含量;流式细胞术检测细胞凋亡率。结果镉染毒内皮细胞损伤后NO浓度明显低于正常对照组,sICAM-1和CytC含量明显升高,细胞凋亡率明显增加,且呈剂量依赖性效应;同时补充硫酸锌处理后NO值增加,sICAM-1和CytC含量下降,细胞凋亡率与正常对照组差异无统计学意义(P>0.05)。结论补锌可通过改善内皮细胞功能,抑制内皮细胞凋亡,达到保护内皮细胞的目的。
Objective To study the anti-injury effects of zinc sulfate on the human umbilical vein endothelial cell(HUVECS)injured by cadmium poisoning and its mechanism. Methods Cultured HUVECS were divided into normal group(NG), cadmium-poisoning group(Cd)and group Zinc-pretreated(Zn), which were respectively cultured with normal serum, cadmium-poisoning serum or with cadmium-poisoning and zinc sulfate pretreated serum for 48 hours. NO concentrations in the culture supernatant were measured by nitrate reductase method, sICAM-1 and Cyt C were measured by ELISA, and flow cytometry(FCM)was used to detect apootosis rate. Results The NO concentration in Cd group was lower than that in NG, the concentration of sICAM-1 and Cyt C were obviously higher than those in NG respectively, the apoptosis rate were increased, and these effects were dose dependent. When pretreated by zinc sulfate,the NO was increased,the sICAM-1 and Cyt C was decreased. In Cd group, the apoptosis were decreased. There was not significanty differences between Zn group and NG. Conclusion The zinc supplement has the protective effect on Cd-poisoning damage of HUVECS. The mechanism may be related to the improvement of vascular endothelial function and inhibition of apoptosis.
