Expression of Twist,E-cadherin and β-catenin in colorectal carcinoma

摘要 目的探讨Twist、E-钙黏蛋白和β-连环蛋白在结直肠癌组织中的表达与患者临床病理指标的关系。方法采用免疫组织化学检测60例结直肠癌和20例同期癌旁正常肠黏膜组织中Twist、E-钙黏蛋白和β-连环蛋白的表达。结果 Twist在结直肠癌组织中的阳性率53.3%显著高于癌旁正常黏膜组织0%;E-钙黏蛋白在结直肠癌组织中的阳性率35.0%显著低于癌旁正常黏膜组织100%;β-连环蛋白在结直肠癌组织中的阳性率18.3%显著低于癌旁正常黏膜组织100%。Twist、E-钙黏蛋白、β-连环蛋白在结直肠癌组织异常表达均与分化程度、浸润深度、淋巴结转移、TNM分期有关,与年龄、性别及肿瘤大小无关。Twist与E-钙黏蛋白和β-连环蛋白呈显著负相关。结论 Twist、E-钙黏蛋白和β-连环蛋白可能协同参与了结直肠癌的早期转移等过程,可能作为反映结直肠癌早期转移的重要生物学指标,Twist将来可能成为结直肠癌靶向治疗的一个新靶点。 Objective To investigate the expression of Twist,E-cadherin and β-catenin in colorectal carcinoma tissue and its association with pathological features.Methods The expression of Twist,E-cadherin and β-catenin in 60 cases of colorectal carcinoma and 20 cases of paracancerous normal tissues was detected by using immunohistochemistry.Results The positive rate of Twist in colorectal carcinoma(53.3%) was significantly higher than that in paracancerous normal tissue(0%).The positive rate of E-cadherin in colorectal carcinoma(35.0%) was significantly lower than that in paracancerous normal tissue(100%).The positive rate of β-catenin in colorectal carcinoma(18.3%) was significantly lower than that in paracancerous normal tissue(100%).The aberrant expression of Twist,E-cadherin and β-catenin was significantly correlated with tumor differentiation,depth of invasion,lymph node metastasis,TNM staging(P0.05),but not correlated with age,gender and size of the tumor(P0.05).Expression of Twist in colorectal carcinomas tissue was negatively correlated with E-cadherin and β-catenin(P0.01).Conclusions Expression of Twist,E-cadherin and β-catenin might play a cooperative role in the onset of malignant progression of colorectal carcinomas.Twist might be a new prognostic marker of malignancy and a potential therapeutic target in colorectal cancer.
