摘要
目的:探讨毒素清对肺炎痰热证大鼠肺组织炎症细胞因子的影响,以揭示毒素清治疗肺炎痰热证的作用机制。方法:制备细菌性肺炎痰热证模型,将Wistar大鼠随机分为正常组、肺炎组、肺炎痰热证组、阳性对照组、毒素清组。采用免疫组化法测定各组肺组织白介素(IL)-1β,肿瘤坏死因子(TNF)-α,白介素(IL)-6,白介素(IL)-10蛋白表达,采用RT-PCR法测定肺组织IL-1βmRNA,IL-10 mRNA表达水平。结果:与正常组比较,肺炎组、肺炎痰热证组肺组织IL-1β,TNF-α,IL-6,IL-10蛋白及IL-1βmRNA和IL-10 mRNA表达水平均显著升高(P<0.01),其中,肺炎痰热证组TNF-α,IL-6,IL-10蛋白及IL-10mRNA较肺炎组升高明显(P<0.01,P<0.05);与肺炎痰热证组相比,毒素清组、阳性对照组肺组织IL-1β,TNF-α,IL-6蛋白及IL-1βmRNA表达显著减弱(P<0.01),其中,毒素清组较阳性对照组减弱明显(P<0.05);与肺炎痰热证组相比,毒素清组肺组织IL-10蛋白及mRNA表达增强(P<0.05)。结论:促炎症因子与抗炎症因子之间的失衡参与了肺炎痰热证的发生发展,抑制促炎症因子IL-1β,TNF-α,IL-6的表达及促进抗炎症因子IL-10的表达可能是毒素清减轻细菌性肺炎痰热证肺组织损伤的主要途径。
Objective:To investigate influence of Dusuqing(DSQ),a traditional Chinese medicine prescip-tion,on expression of inflammatory factor in lung tissue of rat models with bacterial pneumonia and phlegm-heat syn-drome and to reveal the mechanism of DSQ in treatment of the disease.Method:Wistar rats were randomized into five groups:normal,pneumonia,pneumonia with phlegm-heat syndrome,positive control and DSQ treated groups.Rat models with bacterial pneumonia accompanied by Phlegm-heat syndrome were made.The expressions of IL-1β,TNF-α,IL-6 and IL-10 proteins in lung tissue were analyzed by immunohistochemistry staining.The expression of IL-1βmRNA,IL-10mRNA was measured by RT-PCR.Result:The expressions of IL-1β,TNF-α,IL-6,IL-10,IL-1βmRNA and IL-10mRNA in pneumonia group and pneumonia with phlegm-heat syndrome group were markedly higher than those in normal control group(P 〈 0.01).And in pneumonia with phlegm-heat syndrome group the ex-pressions of TNF-α,IL-6,IL-10 proteins and IL-10 mRNA were significantly higher than those in the pneumonia group(P 〈 0.01,P 〈 0.05).Compared with pneumonia with phlegm-heat syndrome group,the expressions of IL-1β,TNF-α,IL-6 and IL-1β mRNA were markedly weakened in positive control group and DSQ treated group(P 〈 0.01) and the expressions in DSQ treated group were weaked significantly than those in positive control group(P 〈 0.05).Compared with pneumonia with phlegm-heat syndrome group,the expressions of IL-10 protein and mRNA were en-hanced in DSQ treated group(P 〈 0.05).Conclusion:The unbalance between proinflammatory and anti-inflammato-ry factor is involved in the pathological damage of lung tissues in pneumonia with phlegm-heat syndrome.The mecha-nism of DSQ in treating bacterial pneumonia with phlegm-heat syndrome may be related to its ability of inhibiting ex-cessive release of proinflammatory cytokine and promoting IL-10 secretion to regulate the balance between pro-and anti-inflammatory responses.
