摘要
目的建立HPLC法测定桑枝、桑叶和桑白皮及其各配方颗粒中桑皮苷A的含量。方法色谱柱为Ulti-mate XB-C18色谱柱(4.6 mm×250 mm,5μm),柱温20℃,流速1.0 mL.min-1,检测波长330 nm,流动相为甲醇(A)-水(B),梯度程序为0~10 min,5%~30%A;10~20 min,30%~40%A;25~35 min,40%~50%A;35~50 min,50%A。结果桑皮苷在1.49~234μg.mL-1质量浓度与峰面积线性关系良好(r=0.999 4),平均加样回收率为100.8%(RSD为1.6%)。桑皮苷A在各样品中含量差异较大。结论本方法准确、可靠、重现性好,可为桑枝、桑叶和桑白皮及其配方颗粒的质量控制研究提供参考。
Objective To determine mulberroside A in Ramulus mori.,Folium mori.,Cortex mori.and their granules by HPLC.Methods Ultimate XB-C18 column was used at 20 ℃.The flow rate and detective wavelength were 1.0 mL·min-1 and 330 nm.The mobile phase was composed of methaol(A) and water(B):0-10 min,5%-30% A,10-20 min,30%-40% A,25-35 min,40%-50% A,35-50 min,50% A.Results The linear range of calibration curve was 1.49-234 μg·mL-1(r=0.999 4).The average recovery of the method was 100.8% and RSD was 1.6%.Conclusion The method is accurate,reliable and reproducible,which can be applied in the quality control of Ramulus mori.,Folium mori.,Cortex mori.,and their granules.
