摘要
为提高重组复合α干扰素的活性和半衰期,在现有复合α干扰素结构基础上进行氨基酸替换,合成了新型干扰素(N-cIFN)基因,将该基因克隆到温控表达质粒pBV220,转化大肠杆菌BL21获得工程菌BL21/pBV220-cIFN,经过诱导表达、包涵体变性、DEAE Sephadex A-25柱层析、Sephadex G-50柱层析以及离心超滤等分离纯化过程,获得纯度为92%的重组人复合α干扰素,对其效价测定结果显示其体外活性为9×108IU/mg。
The gene encoding recombinant human consensus interferon α protein（cIFN） was inserted into the temperature controlled plasmid pBV220.The recombinant cIFN was expressed as inclusion body and purified using DEAE Sephadex A-25,Sephadex G-50 and ultrafiltration device.The purity of cIFN is about 92%.The result of VSV-WISH showed that the bioactivity of cIFN was about 9×108IU/mg.
