摘要
目的:建立高效液相色谱法(HPLC),测定不同批次益母草药材中盐酸水苏碱的含量。方法:采用磺酸基团键合硅胶的阳离子交换剂为填充剂;以0.05mol/L磷酸二氢钾溶液-磷酸-三乙胺(1000:1.5:0.4)为流动相;检测波长为192nm,流速为1.0ml/min,柱温25~C。益母草药材经适当提取纯化。结果:盐酸水苏碱在进样量为2.0996~18.8964μg范围内具有良好的线性关系,r=0.9997;盐酸水苏碱的平均回收率为98.62%,RSD为1.41%(n=5)。结论:本方法专属性强,重现性好,可作为益母草中盐酸水苏碱的定量测定方法。
Objective: To establish a HPLC method for the separation and determination of stachydrine hydrochlofide in leonurus japonicus houtt from different districts. Method: ?The Analytical column was Waters Spherisorb SCX column (250× 4.6mm,5μm).The mobile phase was composed of O.05mol/L KH2PO4-H3PO4-(C2H5)3N (1000:1.5:0.4); UV detection was performed at 192 nm, the flow rate wes 1.0mL/min, column temperature was at 25℃ .The test solution was obtained by sample extracted with ethanol and passed through an actined charcoal-aluminium oxide column using ethanol as eluant. Results: The calibration curve was linear over the range between 2.0996 and 18.8964ug of stachydrine (r=0.9997);the average recover was 98.62 % and RSD was 1.41% (n=5). Conclusion: This method is selective and reproducible, and can be used to determine stachydrine hydrechloride in leonurus japonicus houtt.
