不同浓度的瘦素在胚胎着床中对细胞间粘附分子-1和金属蛋白酶9的影响 被引量:2

The affection of leptin on the expressions of intercellular adhesion molecule-1 and matrix metalloproteinase 9 in the embryo implantation

摘要 目的建立体外子宫内膜培养模型,观察不同浓度瘦素(即leptin)对与着床密切相关的细胞间粘附分子-1(Intercellular Adhesion Molecule-1,即ICAM-1)、金属蛋白酶9(Matrix Metalloproteinase9,即MMP9)的影响,进而探讨leptin对胚胎早期着床的的调节作用。方法采用免疫细胞化学方法检测子宫内膜中ICAM-1和MMP9的表达;采用ELISA方法分别检测经leptin干预培养模型后培养液中ICAM-1和MMP9的表达。结果免疫细胞化学方法:ICAM-1主要在腺体细胞中有表达,基质细胞仅有少量的表达,而MMP9在子宫内膜基质细胞和腺体细胞中均有表达;ELISA结果显示:与对照组相比,ICAM-1经100ng/ml的leptin干预后表达上调(P<0.05),而经1ng/ml、10ng/ml的leptin干预后表达无显著性差异(P>0.05);MMP9分别经1ng/ml、10ng/ml、100ng/ml的leptin干预后表达上调(P<0.05)。结论一定量的leptin可以增加体外培养模型中ICAM-1和MMP9表达。 Objective:In order to study the mechanism of interleukin-1β(IL-1β) and leptin on embryo implantation in the early stage,we have observed the expression of intercellular adhesion molecule-1(ICAM-1) and metalloproteinase 9(MMP9) intervened by leptin which was closely related to the implantation in the vial model of endometrial cells.Methods:The expressions of ICAM-1 and MMP9 in the endometrium were detected by cytohistochemistry.However,the expressions of ICAM-1 and MMP9 in the medium of model which have been intervened seperately by IL-1β,leptin or interleukin-1 receptor antagonist(IL-1ra) were detected by enzyme-linked immunosorbent assay(ELISA).Results:Cytohistochemistry:ICAM-1 was mainly expressed in the glandular cells,rarely in the stromal cells,but MMP9 was expressed in the endometrial stromal cells and glandular cells.The result of ELISA showed that the expression of ICAM-1 in the medium was upregulated after been intervened by leptin(100ng/ml)(P0.05).But after been separately intervened by leptin(1ng/ml,10ng/ml),they had no significant differences compared with the control group(P0.05).The result of ELISA showed that the expression of MMP9 in the medium was upregulated after been intervened by leptin(1ng/ml,10ng/ml,100ng/ml)(P0.05).Conclusion:Certain amounts of leptin can upregulate the expression of ICAM-1 and MMP9.
