摘要
目的:探讨特发性胎儿生长受限(IFGR)患者母血及脐血中胎盘生长因子(PLGF)及胎盘组织中Toll样受体2(TLR2)的表达及意义。方法:选取郑州大学第三附属医院2008年12月2009年10月剖宫产分娩的FGR孕妇28例作为实验组,同期因社会因素剖宫产分娩的正常足月孕妇30例作为对照组。采用双抗体夹心酶联免疫吸附法(ELISA)测定实验组与对照组母血、脐血PLGF浓度;免疫组化SP法检测两组胎盘组织中TLR2的表达(平均灰度值为检测指标)。结果:①IFGR组母血、脐血PLGF浓度与对照组比较,明显降低(P〈0.05);②胎盘组织中均有TLR2蛋白不同程度的阳性表达;③TLR2在胎盘合体滋养细胞上的表达实验组高于正常对照组,差异有统计学意义(P〈0.05);④IFGR组母血中PLGF与胎盘组织中TLR2呈负相关。结论:母血、脐血PLGF减少可能是引起FGR的原因之一。IFGR组胎盘组织中TLR2表达增加可能是IFGR发病的重要因素之一。在IFGR中TLR2可能通过对PLGF的表达下调,抑制胎盘胎儿的发育。
Objective:To explore the expressions and significances of placental growth factor(PLGF) in maternal blood and umbilical cord blood and Toll-like receptor 2(TLR2) in placental tissues in cases with idiopathic fetal growth restriction(IFGR).Methods:28 pregnant women with IFGR who delivered in the hospital from December 2008 to October 2009 were selected as experimental group,30 normal full-term pregnant women who delivered in the hospital because of social factors at the same period were selected as control group;double antibody enzyme-linked immunosorbent assay(ELISA) was used to measure the concentrations of PLGF in maternal blood and umbilical cord blood;the expressions of TLR2 in placental tissues of the two groups were detected by immunohistochemical SP method.Results:The concentrations of PLGF in maternal blood and umbilical cord blood in IFGR group were significantly lower than those in control group(P〈0.05).TLR2 protein expressed in placental tissues to a different extent.The expression level of TLR2 in syncytiotrophoblast cells of placenta was significantly higher than that in control group(P〈0.05).There was a negative correlation between PLGF in maternal blood and TLR2 in placental tissues.Conclusion:The decrease of PLGF in maternal blood and umbilical cord blood may be a cause of IFGR;the increase of TLR2 expression in placental tissues of IFGR group may be an important factor of pathogenesis of IFGR;TLR2 inhibits the development of placenta and fetus by down-regulating PLGF in cases with IFGR.
