三种骨骼肌纤维中碱性成纤维细胞生长因子基因表达的特征 被引量:6

Different expressions of bFGF gene in three kinds of skeletal muscles

摘要 目的 研究碱性成纤维细胞生长因子 (bFGF)基因、bFGF蛋白及其受体在大鼠骨骼肌纤维的表达与分布规律。方法 采用肌纤维特殊染色、免疫组织化学、原位杂交以及受体检测等方法 ,研究bFGF基因、bFGF蛋白及其受体在大鼠后肢腓肠肌的分布。结果 根据特殊染色 ,大鼠后肢腓肠肌纤维可以分成Ⅰ、Ⅱa与Ⅱb三种类型。bFGF基因的阳性表达仅见于Ⅰ型与Ⅱa型纤维胞浆 ,bFGF蛋白及其受体的分布也仅见于Ⅰ型与Ⅱa型肌纤维内膜。结论 本研究首次鉴别出三种不同类型骨骼肌纤维表达bFGF基因、bFGF蛋白及其受体的特征与规律。对深入了解bFGF参与骨骼肌纤维胚胎发生、生长、成熟后表型的维持以及创伤后的修复有重要意义。 Objective To explore the distribution characteristics of bFGF gene, bFGF protein and receptor in rat skeletal muscles Methods bFGF gene, and bFGF protein and receptor in rat gastrocnemius muscles were localized by ABC immunohisto chemical method, in situ hybridization and a special staining methods. Results Based on the results from the special staining, the fibers in rat gastrocnemius muscle could be divided into three types: type Ⅰ, Ⅱa and Ⅱb The positive expression of bFGF gene was only found in the cytoplasm of typeⅠ and typeⅡa fibers The immunohistochemical localization showed that bFGF protein and its receptor were found in the connective parts of muscle endomysium in fibers of typeⅠ and type Ⅱa Conclusion This is the first study to identify the different expressions of bFGF gene, bFGF protein and its receptor in rat skeletal muscles The results will help to understand the action and the relationship between bFGF autocrine and the embryo development of skeletal muscle fibers
