不同脱细胞方法对周围神经生物力学性能的影响 被引量:6

Effects of Different Acellular Methods on Biomechanical Properties of Peripheral Nerve

摘要 探讨新鲜周围神经和经不同脱细胞方法处理的周围神经在生物力学性能上的差异,为组织工程化神经构建提供实验依据。收集17只3月龄雄性Wistar大鼠的双侧坐骨神经,随机分为3组,正常对照组(A组):不经任何处理;物理冻融组(B组):-196℃冷冻和37℃融解处理坐骨神经;化学萃取组(C组):TritonX-200,Sulfobetaine-10(SB-10),Sulfobetaine-16(SB-16)处理坐骨神经。脱细胞过程完成后,3组神经均经HE染色、场发射扫描电镜观察神经结构,后用Endura TEC ELF 3200力学仪器测试3组神经的生物力学性能。HE染色和场发射扫描电镜研究表明:化学方法在去除雪旺细胞、轴突、髓鞘和保留神经结构方面优于物理方法。生物力学测试表明:正常对照组、物理冻融组和化学萃取组的极限载荷、极限应力、极限应变、断裂功耗经统计学检验无统计学意义(P>0.05)。因此,化学方法处理后的神经更适合在移植中应用。
