高脂诱导的NAFLD大鼠模型血清IL-6、TNF-α等的变化 被引量:4

摘要 目的:白细胞介素-6(Interleukin-6I,L-6)和肿瘤坏死因子(Tumor necrosis factor-α,TNF-α)在非酒精性脂肪肝病(Nonalcoholic fatty liver disease,NAFLD)的形成中具有重要作用。我们探讨在大鼠NAFLD发生发展过程中IL-6血清浓度的变化及与TNF-α、胰岛素抵抗(Insulin resistance,IR)的关系,从而更好地了解非酒精性脂肪肝病的发病机理。方法:48只雄性SD大鼠随机分为正常对照组和NAFLD模型组,正常对照组采用普通饲料喂养,NAFLD模型组采用高脂饲料喂养,并于第2、4、6、10周末分批处死各组大鼠6只,并检测其血清ALT、AST、TG、TC、空腹血糖(Fasting blood glucose,FBG)、空腹胰岛素(Fastinginsulin,FINS)I、L-6、TNF-α等指标,计算肝指数及胰岛素抵抗指数(Insulin resistance index,IRI),观察肝脏病理变化。结果:模型组大鼠血清ALT、AST、TG、TC、FINSI、RII、L-6、TNF-α均较对照组升高,其中TNF-α自第4周起高于对照组(P<0.05)I,L-6第6周起高于对照组(P<0.05)I,L-6与TNF-α(r=0.931,P<0.05)I、RI(r=0.946,P<0.05)呈高度正相关。结论:脂肪因子IL-6与TNF-αI、RI密切相关,与NAFLD的形成发展有重要关联。 Objective:IL-6 and TNF-α play critical roles in the development of nonalcoholic fatty liver disease(NAFLD).We thus investigated the changes of plasma Interleukin-6(IL-6) and TNF-α in the nonalcoholic fatty liver of rats and their correlation.This will provide new insight into the pathology of NAFLD.Methods:Forty-eight male SD rats were divided in two groups randomly.The NAFLD was induced in male SD rats by feeding the high-lipid nutrition,whereas,the control group was fed with low-fat nutrition.All rats were sacrificed at the end of 2,4,6,and 10 weeks.The levels of ALT,AST,TG,TC,FBG,FINS,IL-6 and TNF-α in the sera of rats were measured.The liver index and IRI were counted and the pathologic changes of liver were recorded during experiments.Results:Compared with control group,the levels of ALT,AST,TC,TG,FINS,IRI,TNF-α and IL-6 in the sera of NAFLD group all increased significantly(P0.01).The levels of TNF-α in NAFLD group started to increase significantly at week 4(P0.05),while the levels of IL-6 began to elevate from week 6(P0.05).From week 2 to week 10,IL-6 was highly positively related to TNF-α(r = 0.931,P0.05) and IRI(r = 0.946,P0.05).Conclusion:The levels of IL-6 were highly correlated with TNF-α and IRI in the sera of NAFLD mouse models,which indicates that both IL-6 and TNF-α are crucial in the development of NAFLD.
