摘要
目的:研究结缔组织生长因子(Connective tissue growth factor,CTGF)对人胰腺癌细胞(Pancreatic cancer cell-1,PANC-1)生长的影响。方法:采用重组腺病毒介导CTGF(Recombinant adenorirus mecliated CTGFr,Ad-CTGF)感染PANC-1,通过MTT法检测CTGF高表达状态下PANC-1的增殖能力改变,同时通过细胞迁移实验和肿瘤细胞集落形成实验观察PANC-1运动能力和集落形成能力的改变。结果:MTT实验表明rAd-CTGF在人PANC-1中高表达具有明显的生长抑制作用,肿瘤细胞迁移实验和集落形成实验表明rAd-CTGF在人PANC-1中高表达能抑制其迁移能力和集落形成能力。结论:结缔组织生长因子高表达具有抑制PANC-1的作用。
Objective:To explore the impact of connective tissue growth factor(CTGF)on proliferation and migration of human pancreatic cancer PANC-1.Methods:Ability of proliferation and migration were tested after recombinant adenovirus mediated CTGF over-expression by MTT assay,and locomotory capacity and colony-forming capacity were observed by transwell assay and tumor cell colony formation assay in human pancreatic cancer cells PANC-1.Results:High expression of CTGF suppressed the proliferation of human pancreatic cancer cells by MTT assay.The ability of migration and colony forming were suppressed as well when CTGF over expressed by transwell assay and tumor cell colony formation assay.Conclusion:Over expression of connective tissue growth factor(CTGF) might inhibit the proliferation and migration of human pancreatic cancer cell.
