苹果多酚诱导ACC-M细胞凋亡及caspase-3表达的实验研究

摘要 目的:探讨苹果多酚体外诱导人腺样囊性癌ACC-M细胞凋亡的作用机制。方法:MTT法、流式细胞术观察不同浓度苹果多酚对ACC-M细胞凋亡的影响;RT-PCR和Western印迹检测苹果多酚作用下ACC-M细胞caspase-3 mRNA和蛋白表达的变化。应用SPSS13.0软件包对数据进行统计学分析。结果:随着苹果多酚浓度的增加,ACC-M细胞凋亡增多(Ρ<0.05),ACC-M细胞中caspase-3 mRNA和蛋白表达量增加(Ρ<0.05),差异有统计学意义。结论:苹果多酚可体外诱导ACC-M细胞凋亡,激活caspase-3基因mRNA及蛋白的表达可能是其作用机制之一。 PURPOSE:To investigate the inducton of apple polyphenols on the apoptosis of ACC-M cell in vitro.METHODS:The effects of apple polyphenols on the apoptosis of ACC-M cell was observed by 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide(MTT) assay and flow cytometry.RT-PCR assay was used to detect the effects of apple polyphenols on the mRNA expression of caspase-3 in ACC-M cell.At the same time,Western blot was used to observe the influences of apple polyphenols on the protein of caspase-3 in ACC-M cell.The data were analyzed using SPSS13.0 software package.RESULTS:Compared with the control group,apoptosis of ACC-M cell in the experimental group increased and caspase-3 mRNA and protein expression increased significantly with the increase of the concentration of apple polyphenol(Ρ0.05).CONCLUSION:Apple polyphenols could induce cell apoptosis in ACC-M cell in vitro,and one of the mechanisms of apple polyphenols inducing the apoptosis in ACC-M cell was probably to promote the expression of caspase-3 mRNA and its protein.
