摘要
目的:研究DNS比色法测定N-AcGA含量的实验条件,为实际应用中测量N-AcGA的含量提供简便快捷的方法。方法:在DNS实验的基础上,对测定N-AcGA含量的实验反应条件(试剂添加量、显色时间、测定波长以及显色后存放时间)进行了改进。结果:DNS试剂的用量0.5 mL,沸水浴显色时间15 min,显色定容放置15 min后,在500 nm波长条件下测定,数据的准确性和稳定性较好。
Objective: This article modified the experimental conditions of 3,5-Dinitrosalicyclic Acid method and then provided a simple and easy way for the determination of N-Acetyl-D-Glucosamine. Methods: Based on the DNS method, the conditions for determination of N-Acetyl-D-Glucosamine were improved. Conclusion: The results showed that on the conditions of DNS amount around 0.5 mL, boiling bath time not less than 15 minutes, and standing for 15-30 minutes after developing, at the wavelength of 500 nm for measuring, the accurate results can be acquired.
