p53基因在茉莉酸甲酯诱导HepG2细胞凋亡过程中表达变化的实验研究 被引量:3

摘要 目的探讨p53基因在茉莉酸甲酯诱导HepG2细胞凋亡过程中的表达变化.方法 RT-PCR检测HepG2细胞中p53mRNA表达,免疫细胞化学检测HepG2细胞p53蛋白表达.结果 MeJA作用HepG2细胞48h后:p53 mRNA表达水平明显下降(P<0.01);p53蛋白表达水平明显降低(P<0.01).结论 MeJA通过下调mtp53及其蛋白的表达,相对增加了wtp53的比例,使细胞周期相关蛋白的表达发生改变,引起细胞周期阻滞和细胞凋亡,从而发挥抗肝癌作用. Objective To study the expression of p53 gene in HepG2 cellsin apoptosis induced by methyl jasmonate.Method We used RT-PCR to detect the mRNA expression of p53 in HepG2 cells,and used immunocytochemistry method to detect protein expression of p53 in HepG2 cells.Result With the effect of MeJA on HepG2 cells for 48 h:the mRNA expression of p53 was significantly declined(P0.01),the protein expression of p53 was also significantly declined(P0.01).Conclusions MeJA can induce HepG2 cells apoptosis by downregulating the mRNA and protein expression of mtp53,increasing relatively Wtp53 expression proportion,changing the expression of cell cycle-related proteinsm,and then causing cell cycle arrest and apoptosis,which play a role against liver cancer.
