摘要
目的探讨重症肌无力(MG)患者外周血淋巴细胞中差异表达的基因。方法应用基因芯片技术,提取30例MG患者和30名健康对照者外周血淋巴细胞总RNA,采用B IOSTAR Hum an-6-V3型人类全长基因cDNA表达谱芯片进行检测,使用生物统计学及生物信息学方法分析两组之间差异表达基因。结果与正常对照组比较,MG组检测出显著差异表达基因104个,其中表达显著上调的基因44个(比值>5.0),表达显著下调的基因60个(比值<0.2),涉及基因包括免疫、细胞信号传递、代谢、细胞周期、细胞凋亡、原癌与抑癌基因、细胞骨架与蛋白合成等相关基因。结论 MG发病机制涉及众多基因表达的改变,基因芯片技术可快速筛选出与MG发病相关基因。
Objective To investigate the differentially-expressed genes in peripheral blood lymphocyte cells in patients with myasthenia gravis(MG).Methods Using the technique of gene-chip,RNA was extracted from peripheral blood lymphocyte cells in 30 MG patients and 30 healthy controls.Differentially-expressed genes were detected with Biostar Human-6-V3 type human genes of cDNA microarray and analyzed between the two groups by biostatistics and bioinformatics.Results Compared with normal control group,a total of 104 differentially-expressed genes in MG group were screened out.Among them,44 genes were up-regulated expression(Ratio5.0),and the 60 genes were down-regulated expression(Ratio0.2).These genes were involved in the immunity,cell signal transduction,metabolism,cell cycle,apoptosis,oncogenes and tumor suppression genes,cell skeleton and protein synthesis-related genes.Conclusions The pathogenic mechanism of MG is involved the changes of multitude genes expression.The related genes of MG can be selected quickly by the technique of gene-chip.
