美金刚联合丰富环境治疗对快速老化小鼠学习记忆能力及精神行为的影响 被引量:3

摘要 目的探讨美金刚联合丰富环境治疗对快速老化小鼠(SAMP8)学习记忆能力及精神行为的影响。方法 40只6月龄SAMP8随机分为未治疗对照组(对照组)、丰富环境治疗组(丰富环境组)、美金刚治疗组(美金刚组)、美金刚联合丰富环境治疗组(联合治疗组),每组10只。各组给予相应治疗8周后,采用Morris水迷宫试验、自主活动观察及高架十字迷宫试验对小鼠的学习记忆水平及精神行为能力进行测试。结果 (1)Morris水迷宫定位航行试验:丰富环境组、美金刚组及联合治疗组小鼠潜伏期均较对照组明显缩短(均P<0.01),且联合治疗组平均潜伏期较丰富环境组和美金刚组(美金刚组除第4 d以外)明显缩短(均P<0.05);Morris水迷宫空间探索试验:美金刚组和联合治疗组小鼠穿越原平台所在位置的次数较对照组均明显增多(P<0.01);联合治疗组较丰富环境组明显增多(P<0.05)。(2)自主活动观察:丰富环境组、美金刚组和联合治疗组小鼠的自主活动次数较对照组明显增多(均P<0.01);联合治疗组小鼠的自主活动次数较丰富环境组和美金刚组明显增多(均P<0.01)。(3)高架十字迷宫试验:丰富环境组、美金刚组和联合治疗组小鼠在开放臂停留的时间百分比较对照组明显增加(均P<0.01);联合治疗组较丰富环境组和美金刚组明显增加(均P<0.01)。结论美金刚和丰富环境均能提高SAMP8的学习记忆能力及缓解其焦虑抑郁情绪,当两者联合后这些作用更明显。 Objective To investigate the effects of Memantine combined with enriched environment therapy on learning and memory abilities and mental behavior in senescence accelerated mouse(SAMP8).Methods Forty 6-month-old SAMP8 mice were divided randomly into 4 groups: untreated control(control) group,treated with enriched environment(enriched environment) group,treated with Memantine(Memantine) group and treated with Mematine combined with enriched environment(therapeutic alliance) group,10 mice in each group.After 8 weeks of each group was received corresponding treatment,the test of Morris water maze(MWM),Autonomic activities test(AAT) and Elevated-plus maze(EPM) were undergone to evaluated the learning and memory abilities and the mental behavior in mice.Results(1) MWM: compared with control group,in hidden platform trials,the mean escape latency were significantly shorter in enriched environment group,Memantine group and therapeutic alliance group(all P0.01),and which was the shortest in therapeutic alliance group then the enriched environment group and Memantine group(all P0.05).The number of crossing original platform in spatial probe test in Memantine group and therapeutic alliance group were much more than in control group and enriched environment group(P0.01).(2) ATT: compared with control group,the number of autonomic activities in enriched environment group,Menantine group and therapeutic alliance group were significantly increased(all P0.01).(3) EPM: compared with control group,the percentage of time spented in open arms in enriched environment group,Memantine group and therapeutic alliance group were significantly increased(all P0.01),and which in therapeutic alliance group was significantly higher than enriched environment group and Memantine group(all P0.01).Conclusions Memantine and enriched environment can improve the learning and memory abilities and ruduce anxiety and depression in SAMP8.And these effects can be enhanced when Memantine combined with enriched environment.
