摘要
基于亲水相互作用机理,以Polar-Silica色谱柱为固定相,乙腈一磷酸盐缓冲液为流动相,分离尿素、三聚氰胺、三聚氰酸、三聚氰酸一酰胺和三聚氰酸二酰胺。应用STATISTICA软件设计实验,考察不同实验条件,如流动相中乙腈的含量,缓冲盐的浓度,pH以及流速等对分离的影响。结果,三聚氰酸一酰胺和尿素、三聚氰胺和三聚氰酸二酰胺的色谱峰易重叠。前者的分离度会随乙腈含量和缓冲盐浓度的降低而改善,而后者则恰好相反。因此,首次建立了可使尿素等5种化合物实现基线分离的液相色谱条件。
Five compounds,urea,melamine,cyanuric acid,ammelide and ammeline,were separated based on HILIC separation mechanism using a Polar-Silica column as stationary phase and a mixture of acetonitrile and phosphate buffer as mobile phase.STATISTICA was used for the design of experiments to investigate the effects of the factors such as the content of acetonitrile in mobile phase,buffer concentration,pH and flow rate.It was found that the peaks of ammelide and urea as well as those of melamine and ammeline would easily overlap each other.With decrease of acetonitrile content and buffer concentration,the resolution between ammelide and urea became better while that between melamine and ammeline worse.Based on these results,chromatographic conditions for baseline separation of these compounds were first proposed.
