摘要
目的:探讨rOGP对自体微小颗粒骨复合Ⅰ型胶原修复节段性骨缺损的效果。方法:取42只健康大耳白兔,于一侧桡骨制成长1.5cm骨缺损模型,植入200mg自体微小颗粒骨与0.2ml胶原的复合物;随机分为A、B2组,A组每天耳缘静脉注射rOGP 0.5ug/kg;B组每天注射同等剂量的生理盐水:分别于术后8、12周进行骨密度测定:12周进行生物力学检查。另取8只仅加入0.2ml胶原作为空白对照组C组。结果:X线和组织学检查A组成骨效果优于B组。8、12周骨密度测试结果显示A组的骨密度值高于B组。12周的生物力学实验结果显示:3点弯曲实验结果表明A组极限强度值明显高于B组,而且A组的弯曲刚度也好于B组:差异均有显著性意义(P<0.05)。A组因髓腔再通时间早,骨改建塑形更完善,新生板层骨的成熟度亦较高,耐受应力情况也更好。C组则不能产生骨性愈合。结论:本实验显示了全身应用rOGP对自体微小颗粒骨复合Ⅰ型胶原对兔骨缺损有修复作用,并且要优于自体微小颗粒骨复合Ⅰ型胶原的成骨效果。
Objective: To investigate the effect of rOGP to autogenous minute granular hone with I collagen on treatment of segmental bone defects. Methods: The model of 1.Sem long bone defect was created in one radius of 42 Japanese white rabbits. A composite of 200mg antogenons minute granular bone. The rOGP was administered to experimental animals by intravenous injection ( 0.5ug/kg.d ) from the first day after surgery to the day before sacrifiee(groupA). While the another animals were injected with saline solution(groupB). The bone mineral density (BMD) of the defects was evaluated at 8 and 12 weeks. The hiomechanie tests were examined at 12 weeks. In group C, only 0.2ml collagen was implanted as control.Results: Radiological and histological examination revealed that the defects healed completely in both A and B experimental group, hut there were more new bone formation, early bridging and rapid healing in group A compared with those of group B.The value of BMD in group A was higher than that in group B. The three-points bending test of group A showed that ultimate strength and bending stiffness were superior than that of group B, the anti-axial compressive stiffness as well;and the torsion rigidity and maximal torque of group A were also greater than that of group B. There were significantly statistic differences in bone minermal density and biomechanical tests between the two granps. The new lamellar cortical bone formation was more matured, and the remodeling process was more perfect in group A comparing to thet of group B, which might contribute to the better mechanical properties of group A. The defects did not achieve any osseous healing in control group(groupC). Conclusion: It was very effective that systemically administering rOGP to autogenous minute granular bone with I collagen on treatment of hone defects,and supering to autogenous minute gramllar bone with I collagen on treatment of bone defects.
