HPLC法测定抗601合剂中绿原酸的含量

Determination of chlorogenic acid in Kang 601 mixture by HPLC

目的建立抗601合剂中金银花有效成分绿原酸含量测定的方法。方法采用高效液相色谱(HPLC)法,色谱柱为Hy-persil ODS(4.0 mm×125 mm,5μm),流动相为甲醇-水-冰醋酸(15∶85∶0.5),流速1.0 ml.min-1,检测波长为327 nm,柱温27℃。供试品溶液加50%甲醇-水溶液,超声震荡,取上清液。结果绿原酸进样浓度在0.2～2μg范围内与峰面积线性关系良好(r=0.999 8),平均回收率为98.3%,RSD为0.79%(n=6)。结论所用方法准确、简便可行、重现性好,可作为抗601合剂的质量控制方法。

Aim To determine chlorogenic acid as the active ingredient of Flos Lonicerae in Kang 601 Mixture by HPLC.Methods The chromatographic column was Hypersil ODS（4.0 mm×125 nm,5 μm）.The mobile phase was methanol-water-acetic acid（15∶ 85∶ 0.5）.The detection wavelength was set at 327 nm.The temperature was 27℃ with the flow rate of 1.0 ml·min-1.Results The linear range of chlorogenic acid was 0.2～2 μg（r=0.999 8）.The average recovery was 98.3%,and RSD was 0.79%（n=6）.Conclusion The method is simple,accurate and reproducible for quality control of Kang 601 Mixture.
