摘要
目的对比研究荧光原位杂交(fluorescence insitu hybridization,FISH)和显色原位杂交(chromogenic in situ hybridization,CISH)在检测乳腺癌HER2基因扩增状态中的敏感性和准确性。方法对32例经CISH法检测HER2基因状态的乳腺癌标本行回顾性FISH法检测,Kappa检验法对两种结果的一致性进行统计分析。结果 CISH法检测HER2基因状态为高拷贝扩增的10例标本经FISH检测均为阳性;低拷贝扩增的12例标本经FISH检测9例为阳性,3例为阴性;10例无扩增的标本经FISH检测1例为阳性,9例为阴性。两者总体符合率为87.5%,K=0.724,P=0.000。结论 CISH和FISH技术均可用于检测乳腺癌HER2基因状态,但FISH法敏感性和准确性较高。
Objective To compare the sensitivity and accuracy of fluorescent in situ hybridization(FISH) and chromogenic in situ hybridization (CISH) in detection of breast cancer HER2 gene. Methods HER2 gene amplification in 32 samples with breast cancer detected by CISH method was retrospectively tested by FISH method. Kappa test was employed to compare the agreement of the two methods. Results Ten samples with high copy amplification by CISH were all detected to be positive by FISH. Nine of 12 samples with low amplification by CISH were positive by FISH and the remaining 3 samples were negative. Nine of 10 samples with no amplification by CISH were detected to be negative by FISH and the remaining 1 sample was positive. The agreement rate of the two methods was 87.5%, K=0.724, P=0.000. Conclusion Both CISH and FISH technology can be applied to detect breast cancer HER2 gene. FISH method has higher sensitivity and accuracy.
