摘要
目的观察芪莲汤颗粒剂对实验性糖尿病肾病大鼠(diabetic nephropathy,DN)肾组织内核转录因子-κB(nuclear factor-kappa B,NF-κB)、单核细胞趋化蛋白1(monocyte chemoattractant protein-1,MCP-1)表达的影响,探讨芪莲汤颗粒剂对糖尿病肾病大鼠保护作用的作用机制。方法 40只大鼠分为正常对照组、模型对照组、芪莲汤颗粒剂高剂量组、芪莲汤颗粒剂低剂量组。除正常组外其它组大鼠用腹腔注射链脲佐菌素(streptozocin,STZ)联合高糖高脂饲料复制糖尿病肾病动物模型,用芪莲汤颗粒剂干预糖尿病肾病大鼠4个月。实验结束前用代谢笼收集大鼠12h尿液,测定尿蛋白含量;取动物血清测定血清尿素氮(blood urea nitrogen,BUN)和肌酐(creatinine,Cr);取肾脏组织用Westernblot方法检测NF-κB、MCP-1及甘油醛-3-磷酸脱氢酶(glyceraldehy de3phosphate dehydrogenase,GAPDH)的表达,用多媒体成像分析系统采集图像计算NF-κB、MCP-1与GAPDH的比值。结果模型对照组动物血糖、尿素氮、肌酐、尿蛋白含量较正常对照组高(P<0.01)。芪莲汤颗粒剂干预后各项指标都相应降低,给药组与模型对照组比较有差异(P<0.05或0.01)。模型对照组肾组织内NF-κB/GAPDH、MCP-1/GAPDH表达较正常对照组增高(P<0.01),芪莲汤干预后表达降低(P<0.05)。结论芪莲汤颗粒剂可以降低糖尿病肾病大鼠血糖、尿蛋白含量及血清尿素氮和肌酐,减轻糖尿病肾病大鼠的肾损害,该作用可能与下调NF-κB、MCP-1表达有关。
Objective To investigate the effects of QiLian soup granula on expression of nuclear factor-kappa B (NF-κB) and monocyte chemoattractant protein-1 (MCP-1) in renal tissues of rats with diabetic nephropathy and explore the possible mechanism. Methods Forty rats were randomly divided into four groups, i.e. normal control, model control, high dose of QiLian soup (QLS-H), low dose of QiLian soup (QLS-L). Model rats with diabetic nephropathy were induced by high calorie feeds and streptozotocin (STZ) intraperitoneal injection. QLS-H and QLS-L groups were treated with QiLian soup granula by intragastric administration for four months. Normal control and model control groups were treated with normal saline. At the end of the experiment, urine was collected by metabolic cage to determine the protein contents, the blood urea nitrogen (BUN) and creatinine (SCr) in serum were determined, and nephridial tissues were collected to determine the NF-κB and MCP-1 expression by Western blot. Results QiLian soup granula decreased the levels of urine protein, serum BUN and SCr(P0.05 or 0.01). The NF-κB and MCP-1 expression in renal tissue decreased in QLS-H and QLS-L groups(P0.05) . Conclusion QiLian soup granula can improve the renal function of rats with diabetic nephropathy. The mechanism may relate to the down-regulation of the NF-κB and MCP-1 expression.
