摘要 文章采用生物复合酶法对薯蓣皂素进行提取研究,使黄姜淀粉和纤维素得到充分降解,促进葡萄糖与皂素的分离,从而有利于黄姜皂素的提取。通过正交实验进行试验确定复合酶处理黄姜皂素的工艺,即最适酶解条件为温度55℃,pH值4.0,时间6h,每克黄姜酶用量为0.03g,通过该条件进行试验发现,黄姜皂素收率提高26%。同时该工艺与传统的直接酸水解法相比,用量酸减少了74.7%,废水总量和COD浓度分别下降了64.1%、89.6%,且产品熔点高。该工艺具有作用条件温和,减少污水排放量、提高皂素得率、污染小、节约能源的特点,可显著降低黄姜皂素生产中污水排放造成的环境污染难题,是一项清洁生产工艺,具有较好发展前景。 Application of complex enzyme in extraction of Dioscorea sapogenins was studied.The starch and cellulose in yam were completely hydrolyzed and easily separated for the extraction of diosgenin from glucose catabolizing solution.Technical parameters of the complex enzyme extraction by the orthogonal test were determined,with the optimal parameter for extraction as hydrolysis temperature 55 ℃,pH of solution 4.0,time of enzymatic hydrolysis 6 h and amount of enzymatic hydrolysis 0.03 g per Dioscorea zingiberensis.Results showed that diosgenin extraction ratio of such method was raised by 26%.Compared with the traditional method,acid quantity decreased by 74.7%,wastewater quantity and COD concentration declined by 64.1% and 89.6% respectively,with high melting point for diosgenin product.Results showed that the technology is featured by gentle function condition,higher efficiency,less pollution and energy saving,which would remarkably decrease the sewage quantity and successfully solve environment pollution in yam diosgenin production.The technology is a cleaner process and has a bright prospective.
