偃麦草EST和POD同工酶遗传多样性分析 被引量:1

摘要 【目的】了解32份新疆野生偃麦草资源在蛋白质水平上的遗传差异状况。【方法】采用聚丙烯酰胺凝胶电泳方法(PAGE)对其酯酶(EST)和过氧化物酶(POD)同工酶谱进行分析,依据Jaccard相似系数,采用UPGMA方法进行聚类,判定偃麦草种质材料间的亲源关系及遗传多样性。【结果】32份材料EST酶谱共出现13条酶带,可划分为2个主带区,与EST1区相比,EST2区着色较深、酶活性较强。POD酶谱共出现3条酶带。【结论】与POD相比,EST同工酶活性更高、多态性更好,更能显示出供试偃麦草材料在蛋白质分子水平上的差异性;供试种质资源EST和POD酶谱共出现16条酶带,其中10条酶带出现频率高于50%,但一些材料也存在特征谱带;聚类分析将供试材料分为4大类,其中来源于新疆阿勒泰地区的材料酶谱特征表现出与环境条件具有一定的相关性。 [ Objective] In order to understand genetic diversity of 32 Elytrigia repens materials on protein level. [Method]Analysis of esterase and peroxidase isozyme from 32 materials of Elytrigia repens were carried out with PAGE electrophoresis, based on Jaccard similarity coefficient, cluster analysis was carried out with UPGMA method, genetic relationship and genetic diversity were determined. [Result] 13 enzyme belts emerged for EST isozyme patterns of 32 materials, which could be divided into two main band zone. Compared with EST1 zero, EST2 zero dyed deeply and had higher enzyme activity. 3 enzyme belts emerged for POD isozyme patterns. [ Conclusion] Compared with POD isozyme, EST isozyme had higher enzyme activity and polymorphism, which was better to show genetic difference among these materials on protein level. There were a total of 16 enzyme belts emerged in EST and POD isozyme patterns, among which, the emergence frequency of 10 isozyme bands were higher than 50%, and special belts also existed in certain materials. Based on cluster analysis, 32 materials could be divided into four categories, the enzyme characteristics of material collected from Altai region, Xinjiang, showed certain correlation with environment conditions.
