组织型转谷氨酰胺酶在单侧输尿管梗阻大鼠肾脏中的表达及意义 被引量:2

Expression of tissue transglutaminase in renal interstitial fibrosis after unilateral ureteral obstruction in rats

摘要 目的探讨组织型转谷氨酰胺酶(tTG)在单侧输尿管梗阻(UUO)大鼠肾间质中的表达及其意义。方法雄性Wistar大鼠随机分为假手术组和模型组,采用左侧输尿管结扎术制备肾间质纤维化模型。术后第2周末处死各组大鼠,采用HE和Masson染色观察肾组织病变;采用免疫组化法检测肾间质中tTG和纤维连接蛋白(FN)的表达;采用RT-PCR技术检测tTG mRNA表达。结果 HE显示,模型组大鼠肾间质增宽,炎细胞浸润,纤维组织增多,肾小管明显扩张。Masson染色可见模型组大鼠肾间质胶原明显增多。模型组tTG和FN的基因和蛋白表达均明显高于假手术组(P<0.01),并且tTG的蛋白水平与FN水平呈显著正相关(r=0.734,P<0.05)。结论 tTG可能通过交联细胞外基质蛋白,抵抗降解,参与UUO大鼠肾间质纤维化的发生发展。 Objective To investigate the expression of tissue transglutaminase(tTG) in unilateral ureteral obstruction(UUO) rats.Methods The animal models of unilateral ureteral obstruction(UUO) were used.Wistar rats were randomly divided into the Sham-operated group and the UUO group.The rats were sacrificed at day 14.Histologicalchanges in renal tubular interstitium were observed with HE and Masson staining,and the mRNA and protein levels of tTG and FN were detected by RT-PCR and immunohistochemistry.Results Massive accumulation of collagen in tubulointerstitium leads to renal fibrosis.Compared with the sham group,the expression of tTG and FN were significantly increased(P0.01),and the levels of tTG protein had positive correlation with the levels of FN protein(r=0.734,P0.05).Conclusion tTG may play a important role in the pathogensis of renal tubulointerstitial fibrosis.
