响应面法优化麦胚多糖的醇析工艺 被引量:2

摘要 采用水提醇沉法提取麦胚多糖,将提取液浓缩至合适体积再加入不同浓度的乙醇,利用多糖不溶于乙醇的特性将其沉淀出来,采用苯酚-硫酸法测定醇析物中多糖含量,并用相关性检验分析了醇析物质量和多糖含量的相关性,皮尔逊相关系数r=0.884>rmin=0.468,说明两者之间存在显著的相关性。为了提高麦胚多糖醇析工艺中多糖含量,通过Box-Behnken中心组合设计(CCD)及响应面分析(RSM),以醇析物为响应值建立了预测多糖含量的二次多项数学模型,优化了多糖醇析工艺,为现实生产提供可靠的理论依据。 Wheat germ polysaccharide (WGP) was extracted by the method of water extracting-alcohol precipitating.Wheat germ extract will be concentrated to appropriate volume, then different concentrations of ethanol was added to precipitate polysaccha- ride which was insoluble in it. Phenol-sulfuric acid was used to determinate the content of polysaccharide and according to the correlation test, there was a close correlation between the precipitates and WGP.In order to increase the content of WGP, using precipitate as the response-value, a mathematical model was established by Box-Behnken design center (CCD) and response surface analysis (RSM), The model was optimized alcohol precipitating technology and provides theoretical basis for practical production.
