Scid小鼠的繁育及其骨髓象分析 被引量:1

摘要 应用带过滤罩的透明鼠盒在层流鼠柜内繁育 T、 B 淋巴细胞均缺失的重度联合免疫缺陷( Scid)小鼠获得成功。 Scid 小鼠平均每窝产仔数略低于对照组 B A L b/c 小鼠。 Scid 小鼠体重与对照组差异无显著性,但其胸腺和脾脏重量均显著低于对照组。病理切片示 Scid 小鼠胸腺无皮质结构,髓质保留,主要由类上皮和成纤维细胞构成,缺乏淋巴细胞,脾脏内滤泡呈淋巴细胞脱空状态,基本被网状细胞占据;但脾脏红髓与对照组小鼠比较无明显区别。 Scid 小鼠血象、骨髓象内淋巴细胞比值较对照组明显降低,而中性粒细胞比值明显增高。 Scid 小鼠淋巴细胞形态不同于对照组 B A L b/c 小鼠,其骨髓中的淋巴细胞主要为大颗粒淋巴细胞。 Severe combined immunodeficiency (Scid) mice, deficient for both T and B lymphocytes, had been bred successfully in laminar air flow units. The average litter size was 4.8, slightly smaller than that of BAL b/c mice. Body weights of scid mice were the same as those of the control group of BAL b/c mice. It was also revealed that the thymus and spleen were significantly smaller than those of BAL b/c mice. Scid thymus was found to consist of a rudimentary medulla without cortex and contained few lymphocytes, predominated by epithelioid cells and fibroblasts. Scid spleen follicles were empty, virtually devoid of lymphocyte, occupied by reticular cells. But red pulp of Scid spleen was normal as in BAL b/c mice. Circulating lymphocytes of Scid mice only comprised a small fraction of the total leukocytes of peripheral blood (12%~24%),while neutrophils relatively amounted to 76%~87%. Conversely, in normal BAL b/c mice, lympheral blood, smear of bone marrow showed lymphocytic cells of Scid mice absolutely decrease (16%~17% of nucleated cells in bone marrow), and myelocytic cells relatively increase (59%~65% of nucleated cells in bone marrow). In BAL b/c mice, lymphocytic cells were 45%~46% of nucleated cells in bone marrow. Myelocytic cells were 31%~34% of nucleated cells in bone marrow. It is also seen that the morphology of lymphocytes in Scid mice differ from BAL b/c mice. The lymphocytes of bone marrow cells in Scid mice differ from BAL b/c mice. The lymphocytes of bone marrow cells in Scid mainly were large granular lymphocytes.
