摘要
以乙腈水(30∶70, V/ V)为流动相,氯霉素为内标,采用反相高效液相色谱法测定吗氯贝胺浓度。血吗氯贝胺在碱性条件下用乙醚提取,甲醇溶解残留物,在 004~4.0μg/m l浓度范围内线性关系良好,r= 0.9996,相对平均回收率为 970% ,日内 R S D(% )26~3.5,日间 R S D(% )3.7~4.2,最低检出浓度004μg/m l。本法快速、准确,灵敏度高,专一性强,适用于吗氯贝胺临床监测及科研需要。
To establish high pressure liquid chromatographic (HPLC) method for the determination of moclobemide concentrations, moclobemide in human serum was extracted with ester and separated on YWG C 18 colum with a mobile phase of acetonitrile:water (30∶70,V/V)and chloraphenicol was used as internal standerd. All detections occureed at 240nm. The linearity was excellent over the range of 0.04~4.0μg/ml(r=0.9996). The relative mean recovery of moclobemide was more than 97.0% and intra-day and inter-day RSD(%) were all less than 4.2. The minimum detection limit in serum was 0.04μg/ml.The method reported was rapid,simple, sensitive and of excellent precision and could be used in determination of serum moclobemide concentrations both for therapeutic drug monitoring and scientific studies.
