上海地区不同年龄人群ACE基因多态性的变化

Study of the Age Dependency of the Polymorphism of Angiotensin I-Converting Enzyme Gene in A Chinese Population from Shanghai

目的观察上海地区人群中血管紧张素I转换酶（ACE）基因多态性的年龄变化。方法运用PCR技术对上海地区312例正常人的ACE基因插入（insertion，I）／缺失（deletion，D)（I／D步态进行检测。结果研究人群ACEI／D多态D、I等位基因频率分别为0.45、0.55。基因型频率男女间无显著差异（FisherP=0.315）。DD频率随研究人群的年龄增加而降低，DD型与非DD型有显著差异（Mantal-HaenszelX2=10.04，P=0.002）。结论在进行ACEI/D多态与各种疾病的研究时，年龄匹配是组间比较的重要条件。

Objective The aim of this study was to observe the age dependency of the polymorphism of angiotensin I - converting enzyme gene in a Chinese population from Shanghai. Methods An insertion/deletion (I/D) polymorphism in the ACE I gene was detected by the polymerase chain reaction (PCR) in 312 healthy subjects from Shanghai.Results Our study showed an overall frequency of 45 percent for the D allele and 55 percent for the I allele. The genotype distribution did not differ between males and females (Fisher P=0.315). Yet, a marked progressive decrease in DD frequency with age was observed. The odds ratio was 10.04 for DD vs non - DD (by x2 analysis, P=0.002). Conclusion The findings emphasize the importance of age - matched group comparisons in the disease association study of ACE I/D polymorphism.
