莱菔子素同系物抑制环氧化酶-2表达的作用研究

摘要 目的:研究莱菔子素同系物(SFN)对环氧化酶-2的抑制作用及机制。方法:采用实时定量PCR方法(Real Time-PCR)检测COX-2 mRNA的表达;蛋白印记杂交试验(western blot)法检测p38蛋白表达的影响。结果:5μM~20μM莱菔子素同系物能明显抑制膀胱癌细胞COX-2 mRNA表达、诱导p38蛋白的表达及磷酸化;p38抑制剂SB202190预处理T24细胞1h,能完全阻断SFN对COX-2mRNA的抑制作用。结论:SFN可通过活化p38激酶抑制膀胱癌细胞COX-2 mRNA的表达。 Objective : To study the effect of sulforaphene homologue on the expression of COX - 2 and its mechanism. Methods : The levels of COX - 2 mRNA was determined by quantitative real - time PCR. The expression of p38 and phosphorylated p38 were detected by western blotting analysis. Results: Sulforaphene homologue (5μM - 20μM) significantly suppressed the expression of COX- 2 mRNA, p38 expression and its phosphorylation. Pretreatment with inhibitor SB202190 thoroughly masked the level of COX- 2 mRNA downregulated by sulforaphene homologue. Conclusion: These results suggest sulforaphene homologue suppressed the expression of COX -2 mRNA through activating p38.
