子宫内膜癌组织中PTEN及PI3K的表达及临床意义 被引量:4

Expressions of PTEN and PI3K in endometrial carcinoma tissues and their clinical significances

摘要 目的:应用组织芯片技术探讨PTEN及PI3K表达与子宫内膜癌临床病理特征之间的关系及其表达相关性。方法:收集62例子宫内膜病理标本,应用组织芯片技术及免疫组化法检测PTEN及PI3K的表达。结果:PTEN和PI3K在子宫内膜良、恶性组织中阳性表达率相比,差异有统计学意义;在子宫内膜癌高中分化组与低分化组中阳性表达率相比,差异有统计学意义;在无肌层浸润或浸润深度≤1/2组与浸润深度>1/2组中阳性表达率相比,差异有统计学意义;PI3K在无淋巴结转移的子宫内膜癌中及有转移者中阳性表达率相比,差异有统计学意义;PTEN阳性表达率与PI3K阳性表达率呈负相关(γ=-0.631,P<0.05)。结论:PTEN阳性表达与子宫内膜癌组织学分级及肿瘤侵袭性呈负相关;PI3K阳性表达与子宫内膜癌组织学分级、肿瘤侵袭性及淋巴结转移呈正相关;二者在子宫内膜癌发生和发展中呈拮抗作用,表达呈负相关。组织芯片用于多因素比较研究具有高效率、质控好的优点,是一种可靠、快速、经济的高通量分析工具。监测PTEN和PI3K表达,对判断肿瘤恶性程度有重要意义。 Objective:To explore the expressions of PTEN and PI3K and the relationship with clinicopathological characteristics of endometrial carcinoma by tissue microarray technology.Methods:The pathological specimens of endometrium were collected from 62 cases,tissue microarray technology and immunohistochemical method were used to detect the expressions of PTEN and PI3K.Results:There was significant difference in positive rates of PTEN and PI3K between endometrial benign tissues and endometrial malignant tissues;there was also significant difference in positive rates of PTEN and PI3K between well/moderately differentiated endometrial carcinoma and poorly differentiated endometrial carcinoma;there was also significant difference in positive rates of PTEN and PI3K between endometrial carcinoma without myometrium invasion or the depth of myometrium invasion≤1/2 and endometrial carcinoma with the depth of myometrium invasion1/2;there was significant difference in positive rate of PI3K between endometrial carcinoma without lymph node metastasis and endometrial carcinoma with lymph node metastasis;there was a negative correlation between positive rate of PTEN and positive rate of PI3K(γ=-0.631,P0.05).Conclusion:There is a negative correlation between positive rate of PTEN and histological grades,invasiveness of endometrial carcinoma;there is a positive correlation between positive rate of PI3K and histological grades,invasiveness of endometrial carcinoma,lymph node metastasis;there is a negative correlation between positive rate of PTEN and positive rate of PI3K,showing an antagonistic effect;tissue microarray technology has the advantages of high efficiency and good quality control in multivariate comparative study,which is a reliable,fast and economical high throughput analysis tool;monitoring the expressions of PTEN and PI3K plays an important role in predicting the degree of malignancy of endometrial carcinoma.
