摘要
为明确转基因水稻表达的Bt蛋白沿捕食性食物链的传递及其对生态系统中较高营养级生物的影响,在实验条件下(28±0.5℃,R.H=80%,L:D=12h:12h)以田间优势自然天敌拟环纹豹蛛为对象,以取食转Cry1Ab/Cry1Ac基因水稻汕优63(简称Bt水稻)叶片24h的稻纵卷叶螟幼虫为猎物饲养拟环纹豹蛛,并测定了不同处理时间的稻纵卷叶螟幼虫和拟环纹豹蛛体内Bt蛋白的含量。结果表明:分别处理12h、24h、48h、60h的稻纵卷叶螟幼虫和分别处理2d、4d、6d、8d、10d的拟环纹豹蛛体内均检测到一定含量的Bt蛋白,且拟环纹豹蛛体内的Bt蛋白含量明显高于稻纵卷叶螟,对照组均不含Bt蛋白;处理组蜘蛛的发育历期长于对照组,成蛛体重小于对照组,且均达到极显著差异水平(p<0.01);处理组蜘蛛的存活时间显著长于对照(p<0.05),而蜘蛛的蜕皮次数与对照相比无显著差异(p>0.05)。可见转基因水稻表达的Bt蛋白能沿食物链经靶标害虫稻纵卷叶螟传递给蜘蛛,并对蜘蛛的生长发育产生一定影响。
In order to appraise Bt protein expressed by transgenic rice transfer and affect the third trophie level creature on the food chain, The natural enemy wolf spider (Pardosa pseudoannulata) , which dominates paddy field, was used in this experiment(28±0.5℃ ,R. H = 80 % ,L:D = 12h:12h). Meanwhile, Bt content of the larvae of Rice case worm ( Cnaphalocrocis medinalis) and wolf spider in different treating time was quantified. The results showed that a certain amount of Bt protein was measured from Rice case worm treated for 12 h,24 h,48 h,60 h and spiders for 2 d,4 d ,6 d ,8 d, 10 d,respeetively,and the Bt content of spider was more than that of Rice ease worm, but control group had no Bt trace; significantly higher development and lower mature spiders weight were observed in the wolf spider fed on Bt rice compared with control treatment(p 〈 0.01 ); longer spiders life-span than that of management group(p 〈 0.05) though the spiders molting has no significant difference (p 〉 0.05 ). These results demonstrated the Bt protein expressed by transgenic rice transfer to the third trophlc level on food chain, while these protein affect wolf spiders development in a certain extent.
