摘要
目的:探讨柚皮素对气道黏液高分泌的作用和相关分子机制。方法:培养A549细胞株,构建气道黏液高分泌模型,以HNE为刺激因素,柚皮素和DMTU为干预因素,观察细胞内ROS含量、EGFR、P-EGFR及MUC 5AC的表达变化。结果:ROS含量的变化与HNE浓度成正比,柚皮素预处理后ROS明显减少(P<0.01);50nmol/L的HNE组中,EGFR mRNA、MUC5AC mRNA和蛋白及P-EGFR蛋白均较对照组显著增加(P<0.05);各干预组中上述指标均下调(P<0.05),差异均有统计学意义。结论:柚皮素具有抑制气道黏液高分泌的作用。
Objective:To investigate the effects of Naringenin on airway mucous hypersecretion and the relative molecular mechanism.Methods:The cell model of mucous hypersecretion was made by culturing A549 cell lines.Results:A dose-dependent increasing of ROS production in cells exposed to HNE was detected,after the cells were pre-treated with Naringenin,the content of ROS,EGFR mRNA,MUC5AC mRNA and P-EGFR proteins were decreased significantly as compared with the 50nmol/L HNE group(P0.01).After the cells were pre-cultured with Naringenin,DMTU and Naringenin + DMTU,respectively,The above measurements were decreased significantly as compared with the HNE group(P0.01).Conclusions:Naringenin attenuated airway mucous hypersecretion under inflammation.
