两种方法同步检测禽样品沙门菌结果分析 被引量:2

摘要 目的:应用LAMP(Loop-Mediated Isothermal Amplification)快诊技术及国标(GB)传统分离方法对杭州市不同来源75份新鲜禽(鸡)样品进行沙门菌检测,并对结果进行分析。方法:在应用GB法对禽样品进行传统分离、生化及血清学鉴定的同时,利用LAMP快诊技术同步检测沙门菌属invA基因及O9群抗原因子基因,并对两种方法检出率及效率进行比较。结果:GB法检出3份阳性,阳性率4.00%;LAMP快诊检测阳性结果因不同增菌液其检测结果不同,检测阳性率最高达17.33%。结论:禽样品可能受沙门菌污染量较低,以致GB法检出率相对较低,但LAMP技术的检测敏感性明显偏高,两种方法同步应用能全面、合理、科学地反映食品中目标菌的污染程度,建议基层实验人员尝试使用。 Objective:To compare the sensitivity and accuracy of LAMP(Loop-Mediated Isothermal Amplification) and GB methods for Salmonella in raw fowls,seventy-five raw samples from different source were tested.Methods: Both GB method and LAMP method for invA gene and O9 group antigen gene of Salmonella were tested simultaneously,and the sensitivity and accuracy of these two methods were compared.Results: Three samples were Salmonella positive tested by using GB method and the positive rate was 4.00%.Salmonella were found in raw fowls by using LAMP,the positive rates varied in the different enrichment broth,and the highest positive rate was 17.33%.Conclusion: The contamination of Salmonella in these raw fowls maybe low and it was difficult to be detected by using GB method.However,the sensitivity of LAMP was high,and the contamination of the target pathogens in food could be detected really by using these two methods simultaneously.It is recommended to be used in base units for food inspection.
