BnGID1基因干扰载体的构建和对甘蓝型油菜的转化

Construction of BnGID1 RNAi vector and transformation on Brassica napus L.

摘要 BnGID1基因是本实验室克隆到的AtGID1基因在甘蓝型油菜(Brassica nupus L.)中的同源基因.为了验证该基因在甘蓝型油菜中的功能,我们利用pFGC5941双元载体构建了BnGID1基因的干扰载体并将其转化入甘蓝型油菜野生型"3529"中,得到了BnGID1基因表达被部分抑制的转基因植株.性状观察证明,在BnGID1基因表达被部分抑制后,植株表现出明显的接化特征,高度显著降低,叶片产生皱缩.与水稻,拟南芥中的GID1基因缺失突变体的性状比较证明BnGID1基因具有与OsGID1,AtGID1等基因相似的功能. BnGID1 is the homologous gene of AtGID1 in Brassica napus L. To prove its function, a BnGID1 RNAi vector from pFGC5941 vector was constructed and transformed on Brassica napus wild type "3529" to get the transgenic plant with BnGID1 silencing. The observation result indicated the BnGID1 silencing transgenic plant showed the dwarf phenotype., their height lowed and the leaves shrunk. Compared with the mutant of loss-of-function of GID1 in rice and arabidopsis showed the BnGID1 had the same function as OsGID1 and AtGIDls.
