摘要
目的:建立滋肾通关胶囊的高效液相色谱(HPLC)指纹图谱,用于评价其提取工艺的优劣。方法:色谱柱为Kromasil C18(250mm×4.6mm,5μm),流动相为乙腈-33mmol·L-1磷酸二氢钾缓冲液(梯度洗脱),检测波长为317nm,柱温为25℃,流速为1mL·min-1,进样量为10μL。用盐酸小檗碱、芒果苷、桂皮醛3种对照品为参照物,建立混合药材的指纹图谱,并以此为标准图谱指认了供试品中对应的成分。结果:在建立的色谱条件下,样品中的成分得到了较好的分离。以70%的乙醇为提取溶剂优于其他2种溶剂。结论:在滋肾通关胶囊的提取工艺中,指纹图谱的多成分评价方法优于常规的单一指标评价方法。
OBJECTIVE: To establish the HPLC fingerprint of Zishen tongguan capsule and evaluate the pros and cons of ex- traction process. METHODS: The analytical column was Kromasil C18(250 mm×4.6 mm, 5 μm) column with acetonitrile-33 mmol. L-1 monopotassium phosphate buffer solution ,(gradient elution) as mobile phase at flow rate of 1 mL·min-1. The detection wave- length was set at 317 nm and the column temperature was 25 ℃. The injection volume was 10μL using berberine hydrochloride, mangiferin and cinnamaldehyde as control. The HPLC fingerprint of mixed medicine was established and uses as standard chromato- grams. The corresponding composition of samples had been identified. RESULTS: Chemical compositions of samples had been sepa- rated successfully by established method. 70% ethanol was better than other two solvent for extracting. CONCLUSION: The multi-component evaluation method of HPLC fingerprint is better than the normal single-component evaluation method in the study of extraction process of Zishen tongguan capsule.
