摘要
以银莓当年生枝的顶芽和腋芽为外植体,以MS为基本培养基,添加不同配比的植物生长调节剂进行离体培养。结果表明:芽诱导最适宜的培养基为MS+6-BA3.0 mg.L-1+NAA3.0 mg.L-1+蔗糖30 g.L-1+琼脂4.5 g.L-1,增殖培养基为MS+6-BA1.0 mg.L-1+NAA1.0 mg.L-1+蔗糖30 g.L-1+琼脂4.5 g.L-1,生根培养基为1/2MS+NAA1.0 mg.L-1+蔗糖20 g.L-1+琼脂4.5 g.L-1。以2份腐叶土与1份炉灰混合作为基质,移栽成活率可达85%。
We used the terminal bud and axillary bud of Elaeagnus commutata in current year as the explant,and MS as basic culture medium with different combinations of plant growth regulators.The result showed that the best culture medium for bud induction was MS + 6-BA3.0 mg ·L-1+ NAA3.0 mg ·L-1+ saccharose30 g·L-1+ agar4.5 g·L-1,for propagation was MS + 6-BA1.0 mg·L-1+ NAA1.0 mg·L-1+ saccharose30 g·L-1 + agar4.5 g·L-1 and for rooting was 1/2MS + NAA1.0 mg·L-1 + saccharose20 g·L-1 + agar4.5 g·L-1.The survival rate of transplantation was 85% if a mixture of rotten leaf soil: furnace ash(21) was used as soil substrate.
