摘要
目的建立高效液相色谱法同时测定黄龙颗粒中3种成分的含量。方法采用Hypersil ODS-C18柱(250 mm×4.6 mm,5μm),甲醇-0.05%磷酸(85:15)为流动相,流速:0.8 ml/min,柱温:室温,检测波长:254 nm。结果大黄酸、大黄素和大黄酚在5.55-90.28μg/ml、5.24-87.54μg/ml和6.33-95.78μg/ml范围内呈良好的线性关系;相关系数分别0.9998、0.9999和0.9999;大黄酸、大黄素和大黄酚的平均加样回收率为98.17%(RSD=1.98%)、98.83%(RSD=2.05%)和96.81%(RSD=0.73%)。结论该方法简便、可靠、准确,可用于黄龙颗粒的质量控制。
Objective To establish a HPLC method for the simultaneous determination of Rhein,Emodin and Chrysophanol in the Huanglong particle.Methods The determination was performed on Hypersil ODS-C18 column(250 mm×4.6 mm,5 μm)with mobile phase consisted of methanol-0.05%phosphoric acid(85:15)at a flow rate of 0.8 ml/min.The UV detection wavelength was set at 254 nm.Results A good linearity was found over the concentration range of 5.55~90.28 μg/ml(Rhein)、5.24~87.54 μg/ml(Emodin)and 6.33~95.78 μg/ml(Chrysophanol).The correlation coeficient was 0.9998(Rhein)、0.9999(Emodin)and 0.9999(Chrysophanol)respectively.The average recovery of particle was 98.17%(RSD=1.98%)、98.83%(RSD=2.05%)and 96.81%(RSD=0.73%).Conclusion The determination method is simple,reliable,accurate,and it can be used for quality control of Huanglong particle.
