摘要
为研究多拷贝rolB基因在转基因植物中的表达,对转入多拷贝rolB基因的三倍体毛白杨1年生植株进行了苗期生长、叶绿素含量及光合特性的测定。结果表明:rolB基因多拷贝株系和转Ri质粒三倍体毛白杨的苗高、节间距和地径(其中多拷贝株系863B的地径与未转基因三倍体毛白杨一样)均低于未转基因三倍体毛白杨;rolB基因多拷贝株系与Ri质粒转化株系的苗高、节间距和地径的差异无规律性。rolB基因多拷贝株系和Ri质粒转化株系的叶绿素a、b、和a+b含量及净光合速率均低于未转基因三倍体毛白杨,其中01B组多拷贝株系的叶绿素a、b、和a+b含量均低于其Ri质粒转化株系,而63B组多拷贝株系与其Ri质粒转化株系之间的差异无规律性。rolB基因多拷贝株系与Ri质粒转化株系净光合速率的差异无规律性。对蒸腾速率、气孔导度和胞间CO2浓度进行比较,rolB基因多拷贝株系与未转基因三倍体毛白杨和Ri质粒转化株系差异均为无规律。
Seedlings growth,chlorophyll content and photosynthetic characters of one-year old plants of triploid Populus tomentosa transferred with multicopy rolB gene were tested to study the expression of multicopy rolB gene in transgenic plant.The results showed that seedling height,internode length and ground diameter of multicopy rolB gene plants and Ri plasmid transfer poplar were lower than those of non-transgenic poplar(the ground diameter of multicopy plant 863B was the same as that of non-transgenic poplar).No similar differences in seedling height,internode length,ground diameter were found between multicopy rolB gene plants and Ri plasmid transfer poplar.The contents of chlorophyll a,b,a + b and net photosynthetic rate of multicopy rolB gene plants and Ri plasmid transfermed poplar were lower than those in non-transgenic poplar.The contents of chlorophyll a,b and a + b of 01B were lower than those in Ri plasmid transfer poplar,but no similar difference was found between the 63B multiocpy series and Ri plasmid transfer poplar.Furthermore,the net photosynthetic rate in rolB gene multicopy and Ri plasmid transfer poplar,the transpiration rate,stomatal conductance,intercellular CO2 concentration among rolB gene multicopy strains,non-transgenic poplar and Ri plasmid transfer poplar did not follow a consistent trend.
