蜂胶与杨树胶HPLC指纹图谱鉴别 被引量:10

摘要 目的:采用高效液相色谱法(HPLC)建立区分蜂胶与杨树胶的指纹图谱,为蜂胶质量控制和真假鉴别提供依据。方法:样本经95%乙醇提取,采用HPLC测定。色谱条件:Agilent Eclipse XDB-C18(4.6mm×250mm,5μm),甲醇-0.5%磷酸水为流动相,流速0.6mL/min,检测波长268nm,柱温30℃。结果:8个不同地区蜂胶的HPLC指纹图谱十分相似,与杨树胶表现出完全不同的指纹图谱特征。其中蜂胶中有2个共有峰是杨树胶中没有的特征指纹峰,而3个不同来源的杨树胶中有3个共有峰是蜂胶中没有的特征指纹峰,且这5个特征峰明显,相互间没有干扰,易鉴别。结论:该方法重复性好,准确度高,可用于鉴定蜂胶与杨树胶以及蜂胶中是否掺有杨树胶。 Objective:To establish fingerprints method to differentiate between propolis and poplar tree gum by HPLC.Methods:The HPLC method was adopted to detect the samples extracted with 95% ethanol.The separation was performed on Agilent Eclipse XDB-C18(4.6 mm×250 mm,5 μm) with the mobile phase consisted of methanol and 0.5% H3PO4 at a 0.6 mL/min flow rate,a 268 nm detection wavelength and a temperature of 30 ℃.Results:The fingerprint of propolis collected from different places showed the similar characteristics,but they were different from those of poplar tree gum.The chromatographic finger prints of eight propolis samples from different producing areas have two characteristic peaks,while the poplar tree gums don't have these peaks.However,3 characteristic peaks,not obtained in propolis samples,appear in three poplar tree gum samples from different areas.Furthermore,these five characteristic peaks are distinct and easy to identify,with no interference among each other.Conclusions:The method is both repeat and accurate,which can be used to distinguish poplar tree gum from propolis and to detect the adulteration of poplar tree's gums in propolis.
