摘要
目的体外观察补骨脂素加长波紫外线的光化学疗法对髓系原代白血病细胞的影响。方法抽取初治和复发的髓系白血病患者骨髓液分离单个核细胞,体外培养。设空白对照组,单纯紫外线照射组,单纯补骨脂素组,补骨脂素加长波紫外线组。通过锥虫蓝拒染法测定细胞生长抑制率,流式细胞术测定细胞坏死和凋亡。结果细胞培养24 h后,单纯紫外线照射组,单纯补骨脂素组,补骨脂素加长波紫外线组,对细胞生长均有抑制作用,且可导致细胞坏死和凋亡的发生,以补骨脂素加长波紫外线组最为显著。结论补骨脂素加长波紫外线的光化学疗法具有诱导髓系原代白血病细胞凋亡和坏死的作用。
Objective To observe the effects of psoralen plus ultraviolet A(PUVA) on primary human myeloid leukemia cells.Methods Myeloid leukemia cells from diagnosis and relapse samples were cultured in vitro and assigned into the control group,single ultraviolet light irradiation group,single psoralen group and PUVA group.The inhibition rates were observed by Trypan blue staining.The cell apoptosis rates and necrosis rates were detected by cytometer with Annexin-VPI.Results After the incubation of Myeloid leukemia cells for 24h,the results showed that either single psoralen therapy or single ultraviolet light irradiation had inhibiting effect on leukemia cells.The inhibiting effect of PUVA on primary leukemic cells was stronger than that of the single psoralen therapy or single ultraviolet light irradiation.In addition,The most effective of cell apoptosis and necrosis was induced by PUVA.Conclusion PUVA could induce apoptosis and necrosis of primary human myeloid leukemia cells.
