摘要
目的通过观察基质金属蛋白酶-2(MMP-2)与低氧诱导因子-1α(HIF-1α)在子宫内膜异位症中的表达,探讨子宫内膜异位症的发病机制及MMP-2与HIF-1α的关系。方法应用免疫组化法检测子宫异位内膜(异位内膜组40例)、子宫在位内膜(在位内膜组40例)及正常内膜(正常内膜组20例)中MMP-2、HIF-1α蛋白的表达水平。结果 MMP-2、HIF-1α的阳性表达分别位于腺上皮细胞的细胞膜和细胞质及细胞核和细胞质,异位内膜组阳性表达高于在位内膜组与正常内膜组(P<0.05),在位内膜组阳性表达高于正常内膜组(P<0.05);MMP-2与HIF-1α表达强度的变化呈正相关(r=0.657,P<0.05)。结论在子宫内膜异位症组织中MMP-2和HIF-1α的表达明显增高,且HIF-1α可能参与调控MMP-2的表达。
Objective To observe the expressions of matrix metalloproteinases(MMP)-2 and hypoxia-inducible factor(HIF)-1α in endometriosis and to study the development mechanism of endometriosis and relationship between HIF-1α and MMP-2.Methods Immunohistochemistry was used to examine the expression of MMP-2 and HIF-1α in 40 cases of ectopic endometrium,40 cases of ectopic endometrial and 20 cases of normal endometrium.Results Positive expression of MMP-2 and HIF-1 were located in the cell membrane and cytoplasm or nucleus and cytoplasm,the positive expression of ectopic endometrium was higher than eutopic endometrium group and normal endometrium group(P〈0.05).The positive expression of eutopic endometrium was higher than normal endometrium(P〈0.05);There were significant positive correlations among changes of expressions in MMP-2 and HIF-1α(r=0.657,P〈0.05).Conclusion The expressions of HIF-1α and MMP-2 were up-regulation in endometriosis.HIF-1α may control the expressions of MMP-2.
